Privileged structures are a class of molecules that are capable of binding to multiple receptors with high affinity. In order to be considered privileged, a substructure should represent a molecule’s core element and make up a significant portion of its total mass. Recent reports^1,2,3 have shown the presence of the arylpiperazine scaffold in potent and selective agonists of the human melanocortin-4 receptor (Fig 1).
