Spent Medium Analysis consists of a battery of complementary tests for nutrient analysis designed to monitor cell growth and provide valuable information and support for cell and media optimization studies. We offer quick turn-around time, with testing typically completed within 10 working days.

88103 Amino Acid Analysis (HPLC)
Performed by Reverse Phase High Performance Liquid Chromatography (HPLC) which is a widely accepted analytical technique that simultaneously separates and identifies individual compounds in complex mixtures. Results can provide valuable information to determine amino acid consumption and formation. Results are reported as a percentage of the fresh media control (qualitative) and actual values (quantitative).

88102 Cholesterol
Cholesterol is a critical nutrient utilized during cell growth. Analysis of this nutrient by HPLC reverse phase can provide valuable information to determine cell consumption of cholesterol. Actual values are reported.

88204 Endotoxin (LAL)
Determined by kinetic chromogenic Limulus Amoebocyte Lysate methodology. Endotoxin is released from the cell membrane of gram negative bacteria. This test method can be used to evaluate spent and fresh media samples, serum and raw materials.

88202 Glucose
Levels are measured by the hexokinase test method. Results provide valuable information to determine spent media glucose consumption during the cycle.

88212 Growth Promotion (MTT)
Evaluates whether a specified cell culture media supports growth of a specified cell line. Growth promotion characteristics are measured by the cellular uptake of tetrazolium salt in an MTT assay. Cells propagated in test samples are compared to cells propagated in an established control. Test requests are evaluated independently based on current capabilities.

88101 Lipid/Fatty Acids
Lipids are extracted, then analyzed by Gas Chromatography Analysis. Lipid classes quantified include phospholipid, free fatty acid, triglycerides, cholesterol esters and total protein. Actual values are reported.

88206 Osmolality
Cellular osmolality (pressure) can impact cell performance. Analysis of osmolality in fresh and spent media can assist in optimizing cell culture media for optimum cell performance. Osmolality is determined by freeze-point depression. Results are measured in mOsm/kg H2O. Tests can be performed with or without sodium bicarbonate.

88207 pH
Cell culture systems require specified pH ranges for optimum cellular performance. The pH of fresh or spent cell culture media is measured with a pH meter. Determination of pH is the negative log10 of the hydrogen ion concentration of the liquid/liquefied samples. pH can be measured with or without sodium bicarbonate.

88211 Solubility
This test determines and documents the solubility of raw materials and dry powder media. Satisfactory solubility is defined as fully soluble in water within 30 minutes at approximately 25 C. The solution is clear and free of particulate. Results are reported as satisfactory or unsatisfactory.

88100 Vitamin Analysis (HPLC)
Water soluble vitamins using reverse phase High Performance Liquid Chromatography (HPLC) is a widely accepted analytical technique that simultaneously separates and identifies individual compounds in complex mixtures. Results can provide valuable information to determine vitamin consumption and formation during cell cycles. Results are reported as a percentage of the fresh media control (qualitative) and actual values (quantitative).

TBD Stability
A custom stability testing protocol is established to meet specific customer testing requirements at specified time points to assess media component degradation. Testing can include but is not limited to pH, osmolality, appearance, vitamin and amino acid analysis. Once a protocol is established and approved, suggested pricing will be established and a standard set-up fee will apply.
