Monoclonal antibodies
produced in mammalian cell culture systems are becoming increasingly important for the treatment of human
disease. Today, therapeutically useful monoclonal antibodies are produced either by hybridoma
cells, or as recombinant proteins in cell lines such as those derived from Chinese Hamster Ovary (CHO) cells. Please see our CHO media web-page for more details.
Traditional techniques for monoclonal antibody production using hybridoma cells have relied upon the use of medium,
such as Iscove's Modified Dulbecco's Medium (IMDM), supplemented with various
components (insulin, transferrin, selenium, pyruvate) and/or bovine sera. In the beginning, monoclonal
antibodies were employed primarily for research or in vitro diagnostic applications, and there was only limited incentive to invest time and money in
the elimination of serum as a supplement.
However, as new technologies have developed, cell culture produced monoclonal antibodies
have become important as potential in vivo human therapeutic agents. These changes in the intended use of monoclonal antibodies have led
to new concerns about the materials and methods employed for monoclonal antibody
production. The threat of contamination of the medium, and ultimately the
final therapeutic drug by adventitious agents extant in animal-derived materials, has led many regulatory agencies to strongly recommend the
discontinued, or limited use of serum and animal-derived materials. In
response to this situation, Sigma has developed several media for the growth and
maintenance of monoclonal antibody producing hybridoma cells which are serum-free, and/or animal-derived protein-free.
The table below lists the serum-free, animal-derived protein-free and chemically-defined
media which Sigma offers for use in the production, by hybridoma cells, of
therapeutically useful monoclonal antibodies. For more traditional media that

Serum-free, low-protein medium formulated to support the growth of hybridoma
cells used in the manufacture of monoclonal antibodies. It supports the rapid growth of cells to high densities, and the maintenance
of viable cells at these high densities for extended periods of time. Hybridoma cells may be transferred without adaptation into this
medium from serum supplemented media.
Because hybridomas can be cultured in this medium without expensive and
chemically complex serum supplementation, down-stream purification is simplified and overall production costs are reduced. |

The purification, of
monoclonal antibodies, which are produced by hybridoma cells, may be simplified
when grown in this chemically defined medium.
This medium is buffered with sodium bicarbonate and MOPS buffer. MOPS buffer has
a pKa = 7.2 with an effective buffering range from 6.5-7.9. It's inclusion in this medium increases buffer capacity and stability within the
optimal pH range when hybridoma cells are being maintained at high densities, and producing monoclonal antibodies.
This formulation eliminates regulatory concerns, about the risks
of biological contamination of recombinant therapeutics, that are associated with the use of serum or animal derived proteins.
