Normal attachment, growth, and development of many cell types are dependent on attachment factors and extracellular matrix components. While some cells are able to synthesize these components, others require an exogenous source, particularly when grown in serum-free culture.

Proteoglycans, which consist of a core protein with one or more
glycosaminoglycan side chains, are a significant component of the extracellular
matrix. Proteoglycans interact with a variety of molecules in the extracellular matrix,
including various cell adhesion molecules and growth factors. To assist in your extracellular
matrix research, Sigma now offers several proteoglycans isolated from different sources.
Aggrecan
Activity:
Combines with hyaluronic acid to form a very large cartilage macromolecular complex.
A 2 mg/ml solution of aggrecan will increase the relative viscosity of a 4% hyaluronic acid solution by 40%.

Biglycan
Activity:
Binds to TGF-b; binds to collagen type I in low ionic strength
(less than 3 mM phosphate) buffer. At higher ionic strengths, biglycan does not bind to collagen
type I. It enhances the inhibition effect of TGF-b on osteoclast proliferation
at a concentration of 4-20 µg/ml. It also prevents the attachment of CHO cells to fibronectin,
with a 50% inhibition at 17-21 µg/ml.

Decorin
Activity:

Heparan Sulfate Proteoglycan (HSPG) Sterile-filtered solution
Activity:
Binds to a variety of molecules found in the extracellular matrix, including laminin, fibronectin,
collagen type IV, and FGF-basic. It induces high affinity binding of FGF-basic to cells deficient in
heparan sulfate and soluble FGF receptors at a concentration of 10-100 ng/ml.

Poly-Lysine
Sigma offers both Poly-D-Lysine and Poly-L-Lysine in several molecular weight
ranges. Poly-Lysine enhances electrostatic interaction between negatively-charged ions of the
cell membrane and positively-charged surface ions of attachment factors on the culture surface.
When adsorbed to the culture surface, it increases the number of positively charged sites
available for cell binding.

Disintegrins
Disintegrins interfere with cell adhesion to the extracellular matrix, including
adhesion of melanoma cells and fibroblasts to fibronectin. They are also potent inhibitors
of platelet aggregation. To meet your research needs, Sigma offers several disintegrins isolated
from different venom sources. These peptides are of low molecular weight, cysteine-rich, and contain
the arginine-glycine-aspartic acid (RGD) sequence. They are all integrin
b1 and b3 inhibitors.

PRODUCT USE

Optimal conditions must be determined for each cell line and
application.
- Add 50 ml of sterile tissue culture grade water to 5 mg of poly-
lysine.
- Aseptically coat culture surface with 1.0 ml/25 cm2 of solution.
Rock gently to ensure even coating of the culture surface.
- After 5 minutes, remove solution by aspiration and thoroughly
rinse surface with sterile tissue culture grade water.
- Allow to dry at least 2 hours before introducing cells andmedium.
*Note: Step 1 is not necessary for Poly-lysine solution, Product
Nos. P 4707 and P 4832.
