Sigma now offers three new Nerve Terminal Stain Kits for the detection and study of synaptic vesicle recycling in neuronal synapses and neuromuscular junctions. Each kit contains a different amphiphilic styryl pyridinium dye for visualization of vesicles with exceptionally low background. The fluorogenic dye differs from kit to kit, although tissue preparation, required reagents and staining procedures are identical. |
Features and Benefits:
- Dyes used include SynaptoGreen C4, SynaptoRed C2 and AM 1-43:
Ideal dyes for fluorescent microscopy applications. Allows intense fluorescence in plasma membrane, but is negligable in aqueous environments.
- Addition of 500 mg of ADVASEP-7 (sulfonated ß-cyclodextrin):
Reduces background and saves time. ADVASEP-7 is applied as a rinse at various stages of the process. The protocol included with each kit reduces excessive washing typically required to remove excess dye from the outer leaflet of the plasma membrane. This process allows only active presynaptic terminals to be labeled with the dye. ADVASEP-7 further reduces background staining by encapsulating the probes, preventing stain from accumulating interstitially.
