Sigma-Aldrich now offers the QuantiGene® RNA Quantification System to measure gene expression knockdowns. As a complement to the MISSION™ shRNA Library, the QuantiGene RNA Quantification System and probe sets provide researchers with the tools to accurately validate RNAi gene knockdown assays. The probe sets and reagent kit are necessary to use the QuantiGene system in validating gene expression.
The QuantiGene® RNA Quantification System is a sandwich nucleic acid hybridization assay that provides a unique approach for RNA detection and quantification by amplifying the reporter signal using branched DNA (bDNA) technology. The bDNA technology allows quantification of target-specific RNA molecules directly from a variety of samples types without RNA purification or target amplification. By measuring RNA directly from crude cell lysates or tissue homogenates, the assay avoids variations or errors inherent to extraction and amplification of target sequences.

Step 1 Lyse cells to release mRNA in the presence of target probes. During overnight incubation, probe set anchors the specific target sequence to the capture plate through multiple CEs binding to the capture plate. |
