The SpyLine Plant mRNA Capture Kits provide a useful protocol for the isolation of RNA from plant tissues and eliminate the measured addition of isolated RNA to a RT-PCR reaction set-up. The elimination of this transfer reduces hands-on time and removes a step where bias and contamination could be introduced. Since the Kits are based on a 96-well PCR plate, the protocol also increases throughput capability and allows for partial automation of purification and RT-PCR set-up. The SpyLine Plant mRNA Capture Kits use oligo d(T) anchored to the wells of a 96-well PCR plate to isolate mRNA from plant cell lysates. This allows RT-PCR to be performed in the same plate that is used for mRNA isolation and eliminates the need to transfer isolated RNA to a secondary vessel for analysis. All kit formats contain a lysis reagent that efficiently releases RNA from partially homogenized plant tissues. After lysis, centrifugation clears cell debris. The resulting supernatant is aliquoted to plate wells and a 30 minute incubation allows the poly (A) RNA contained in the sample to bind to the anchored oligo d(T). The supernatant is then removed and the wells are washed 4 times to ensure complete removal of any contaminants. RT- or qRT-PCR reagents are then added to the plate wells and the entire plate is moved to a 96-well thermal cycler. One- or two-step qRT-PCR may be performed with specific (TaqMan®, Eclipse™) or non-specific (SYBR® Green) reporter chemistries. Optionally, isolated mRNA can be eluted from the SpyLine mRNA Capture Plates.
