Locate shRNA sets for your specific gene of interest through the MISSION shRNA Target Search database. The database may be searched using RefSeq or Accession Numbers, TRC numbers, gene symbols, gene descriptions among other specific identifiers. Target sets found in the collection based on your search criteria will be returned on a Search Results Page and listed by RefSeq number. The results can be further refined and sorted or you may click on a target to obtain in depth information about the shRNA set. Please refer to the help files in the event you have trouble using the database or locating your target.
The library is currently provided as frozen bacterial glycerol stocks harboring sequence-verified shRNA lentiviral plasmid vectors or as purified plasmid DNA. Each gene is represented by a target set that consists of an average of 3-5 individual constructs or clones targeting different regions of the gene sequence. Bacterial cultures may be amplified from the glycerol stocks for use in purification of the shRNA plasmid DNA. Alternatively, the purified plasmid format does not require these extra steps as the growth and DNA purification are completed for you. Target cell lines may be transfected with the purified plasmid for transient or stable gene silencing (puromycin selection). In addition, self-inactivating replication incompetent lentiviral particles can be produced in packaging cells (HEK 293T) by co-transfection with compatible packaging plasmids.
Orders of 5 or less target sets (≤ 25 constructs) are provided in individual 1.4 ml TrakMates® tubes. Each TrakMates® tube contains a unique 2 dimensional barcode on the bottom of the tube that can be read using a corresponding reader. A printed value corresponding to the The RNAi Consortium (TRC) clone number is also provided on each tube. Orders of >5 clone sets (>25 constructs) are automatically provided in a 96-well plate with a one dimensional barcode label on the plate and are provided with a CD containing plate map positions.
