Do you guarantee knockdown using your clones?
Based upon the design algorithm developed by The Broad Institute, 1 of 5 clones is predicted to give greater than 70% knockdown of the targeted transcript. We are currently validating the library and for the target sets we’ve tested, this holds true. Generally more than one clone in each set gives significant knockdown.

Have you validated all of the target sets?
We are in the process of validating the entire library. As data is generated, we will provide the results on our website.

What format can the shRNA clones be ordered?
Target sets consisting of 3-5 clones are available for 7,100 human and 2,900 mouse genes. They may be ordered as complete target sets or individual clones. 3 formats are available for each target set/clone: bacterial glycerol stock, purified plasmid DNA, and lentiviral particles.

What does it mean if I cannot find my target gene in the database?
The collection is currently being cloned. New target genes will be introduced on a quarterly basis until the library is completed. It is possible your target gene is not in the current collection, but may be coming in future releases. Also, it is possible that alternate accession numbers or gene symbols may be used for your gene of interest. Please check NCBI for up-to-date information.

What gene family sets are available?
The kinase panel is currently available. Additional sets will be made available over the coming months. If your gene set of interest is not yet available, please inquire for advanced custom preparation at RNAi@sial.com

How should I store my bacterial cultures?
Glycerol stocks should be maintained at -80°C. Cultures should be maintained fresh, avoiding prolonged storage at 4°C.

What if I have trouble obtaining high yields of plasmid DNA?
Viral-based vectors are known to be somewhat difficult to prep. If particular clones are troublesome, we recommend GenElute™ HP Midi or Maxi prep kits.

Are there special protocols for transfection of the shRNA plasmids?
No. You may use your current method of transfecting plasmid DNA. If you do not currently transfect plasmid DNA, we recommend ESCORT™ V Transfection Reagent.

What is the process for ordering MISSION shRNA clones?
Due to our LIMS system tracking and management of the clone collection, these products may only be ordered online. First, create an online profile and request order access (one time step that takes 24-48 hours for approval). Login to the Sigma-Aldrich Web site, navigate to the MISSION target search database, add target sets to your order cart, and checkout referencing you PO# or credit card number. Click on the ordering tutorial located on the target search Web page for more detailed information.

How do I create an online profile with order access?
At the top of the Web page, select “Your Profile”, follow the instructions and complete all required fields. Be sure to select the box indicating that you are requesting order access and complete any additional required fields.

Is customization of large format orders possible?
