Synthesis Scales Our synthesis scales include 0.03 (3 O.D. Select), 0.05, 0.2, and 1.0 µmole. Large scale synthesis is available upon request.
Purification All Sigma-Genosys oligos include reverse-phase cartridge (RP1)at no additional charge. For stringent applications, additional purification is available. Purification options include desalting and deprotection (no charge), reverse-phase HPLC, and PAGE. If you are unsure about the purification level you need for your research, see our technical bulletin entitled Selecting the Best Purification or contact our Technical Support staff for assistance.
Minimum Yield Yield will vary based upon base composition, purification, oligo length, scale and modifications. That's why we have set minimum yield standards - guaranteed quantity. However, it is typical that you may receive more than the minimum yield guarantee.

Modified Oligos
Sigma-Genosys offers dual-labeled probes for use in applications such as real-time PCR, multiplexing, SNP analysis, allelic discrimination and spectral genotyping. We are a licensed supplier of Molecular Beacons and Black Hole Quenchers.
Capabilities Dual-labeled fluorogenic probes and Molecular Beacons incorporate a 5'-reporter dye and a 3'-quencher within the probe design. Sigma-Genosys can provide the traditional TAMRA quencher for dual-labeled fluorogenic probes and DABCYL quencher for Molecular Beacons.
However, a new family of quenchers is now available that has been optimized for FRET - the Black Hole QuencherTM. Sigma-Genosys is licensed to sell BHQTM labeled oligos for optimum quenching efficiency. Each BHQ has been designed to work with specific fluorophores. BHQ is suitable for both dual-labeled fluorogenic probes and Molecular Beacons.

Dual Label Probes
Capabilities Sigma-Genosys can attach fluorescent dyes to oligos at the 5'-end, 3'-end and even internal positions. Our product offering includes common dye modifiers such as 6-FAM, HEX, and TET, as well as exotic modifications. We are a licensed supplier of dyes from Molecular Probes, Inc., LI-COR, Biosearch Technologies, and Mosaic Technologies.
In addition to standard fluorescent modifications, we have the capabilities to provide dual-labeled probes including Molecular Beacons and Black Hole Quenchers.

Purification Desalt or cartridge (RP1) purification is acceptable for most common phosphoramidite modifiers. However, additional purification such as HPLC is strongly recommended for modifiers that require NHS-ester chemistry to conjugate a dye through an amine linker such as Molecular Probes dyes.
Minimum Yield Due to the chemistry of many modifications, yield will be approximately 40 - 60 % less than a standard oligo. Please see our modification yield chart for minimum yield guarantees.
Quality All modified oligos are quality checked by PAGE and spectrophotometer analysis. In addition, we perform specialized QC methods tailored to specific modifications, such as OD ratio measurement where appropriate.
