Custom DNA Oligos
 

The "Oligonucleotide Technical Data Sheet" is shipped together with all orders and contains all the information related to the oligos we manufactured for you. It can also be provided electronically or on hard disk upon request. This can be particularly useful if you order very large batches of oligos.

Oligonucleotide General Information:

Cat:

GUAR, Easy, FAST, followed by the quantity in OD you ordered. For GUAR oligos, e.g.:

• GUAR5 means you ordered 5 OD of Guaranteed oligo

Batch number:

The number under which all production data related to an oligonucleotide are stored in our production software (very important in case you have technical questions on this oligo)

Type:

Base composition

MP:

Position on microplate (96 well plate oligo orders only)

Purification:

All Sigma-Proligo are systematically desalted. You can order either desalted oligos (non purified) or PAGE or HPLC-purified oligos

Name:

Name of your oligo, as you have supplied

Sequence:

With e-mail and on-line orders, the sequence is electronically sent to the synthesizer from our Production Central Workstation without any typing.

After synthesis, the exact sequence synthesized is printed on the data sheet. The sequence on the data sheet is a 100% guarantee of the sequence synthesised.

Size:

Length of your oligo

Epsilon:

Molar Extinction coefficient value. This coefficient is automatically calculated by the Sigma-Proligo workstation and is based on the exact nucleotide composition of each oligonucleotide.

MW:

Molecular Weight of your oligo, using the values:
A: 313, G: 329, C: 289, T: 304, N: 309

Quantity OD:

OD units are obtained by spectrophotometric measurement at 260 nm. One OD corresponds to the amount of oligonucleotide in a 1 ml volume that results in an optical density of 1 in a 1 cm pathlength cuvette. This corresponds approximately to 33µg of oligonucleotide for a 20mer. However the precise quantity of your oligo is calculated by our station, using both the measured OD value and the calculated Epsilon value.

Quantity nmoles:

Precise quantity of delivered oligo, calculated by our station using measured OD value and calculated Epsilon value

Concentration:

in nmole/µl = total OD x 103 / extinction coefficient x volume (µl)

in ng/µl = concentration (nmole/µl) x MW

Tm:

Melting temperature (non modified oligonucleotide): Temperature at which 50% of the oligonucleotide is hybridized to the complementary strand. For oligonucleotides > 20 bases, calculation is based on the Nearest-Neighbor formula with [salt] = 50 mM and [oligo] = 250 pM. For oligonucleotides = 20 bases we use the Wallace rule Tm = 2(A+T) +4(G+C).
