Expression - Codon usage is engineered for optimal expression in bacterial, yeast, and plant systems, etc.

Genetic manipulation for protein structure and function studies - Appropriate restriction sites can be included in the gene sequence, dividing the gene into modules or cassettes for protein domain swapping/insertion/deletion/duplication.  In addition, specific mutations may be introduced within a particular gene module.
