Sigma-Genosys offers several options for custom antibody production
Antigen Design and Synthesis
- Use of sophisticated software to analyze protein sequence for antigenicity,
flexibility and B-turn formation
- Peptide chemistry analysis is performed on the selected peptide
segment to ensure synthetic manufacturing success
- Peptide is synthesized using proprietary Pepticus synthesis technology
with Fmoc chemistry
- Peptide is analyzed by mass spectrometry and reversed phase HPLC
to confirm composition and purity respectively
Peptide Conjugation
Host Animals
- Two New Zealand white rabbits are standard host animals
- Chicken, Sheep and Goats are available
- Avoid self-recognition of the immunogen by the host animal by selecting
a host species that is very different from your immunogen source.

Immunization & Sera Collection
- Antigen is injected subcutaneously into two host animals
- Initial immunization is given in Complete Freund's Adjuvant and all subsequent immunizations in Incomplete Freund's Adjuvant.
- Pre-immune bleed is drawn and supplied to customer
- 77-day schedule includes 6 immunizations per animal and 4 bleeds per animal
- ELISA is performed on pre-immune and first production bleed
- Sera are supplied unpurified (purification available upon request) and contain several different isotypes (IgG, IgM and IgA)
