What is LNA?

LNA is a novel type of nucleic acid analog that contains a
2'-O, 4'-C methylene bridge. This bridge restricts the flexibility of the ribofuranose ring and locks the structure into a rigid C3-endo conformation, conferring enhanced hybridization performance and exceptional biostability.



LNA® oligos are innovative oligonucleotides that contain Locked Nucleic Acids™ (LNA) to provide superior hybridization characteristics and enhanced biostability compared to conventional DNA oligonucleotides.

LNA-containing oligos are provided as follows:

  • Deprotected and desalted
  • Purified by PAGE, RPC or RP-HPLC upon request
  • Delivered in solution in deionized water or dried down
  • Systematic QC by MALDI-TOF mass spectrometry analysis and - depending upon the nature of the oligonucleotide requested - by RP-HPLC or IE-HPLC

LNA oligos are composed of either 100% Locked Nucleic Acid (LNA) bases or a mixture of LNA and DNA bases. However, only oligonucleotides that are between 7 and 15 mers in length can be made up of 100% LNA. Oligos heavy in LNA content may exhibit problematic self-complementarity. Therefore, for oligonucleotides longer than 15 mers, we recommend decreasing the LNA content as the oligonucleotide increases in length, as follows:

LNA oligos are available at our standard synthesis scales (0.05, 0.2, 1 µmole). LNA bases may also be incorporated into our Real-time qPCR probes. Consult with your local office for guaranteed yield and delivery times.

LNA oligonucleotides can be designed to enhance a wide variety of genomic applications and technologies that rely upon the use of oligonucleotides:

  • Hybridization probes for the detection of nucleic acids and genotyping
  • Padlock probes, modified Taqman™ probes and probes for in situ hybridization
  • Capture probes for target enrichment, sample preparation and affinity chromatography
  • Allele-specific PCR1
  • Antisense oligonucleotide technology
  • Oligonucleotides for therapeutic or diagnostic applications, such as, aptamers, ribozymes, decoy oligonucleotides and siRNA
  • PCR-based applications

In most respects, LNA oligonucleotides can be used in the same manner as DNA oligonucleotides: it is soluble in aqueous buffers, can be ethanol precipitated, dried and resuspended, and analyzed by gel electrophoresis, HPLC and MALDI-TOF.

Every oligo is quality checked by MALDI-TOF mass spectrometry, electrospray ionization mass spectrometry (ESI-MS), capillary electrophoresis (CE), and/or polyacrylamide gel electrophoresis (PAGE).
