Regulatory
agencies worldwide have recently expressed concern about the safety of products,
particularly therapeutic injectables, produced in culture media containing
animal derived components. This heightened concern is due to the potential
fopr contamination by adventitious agents (e.g. TSE's). Manufacturers have
been advised to avoid the use of serum and other animal derived materials in
the manufacturing process. These recommendations are likely to become rigid
requirements over the coming years. For this reason, attention has focused
on the development of media devoid of components of animal origin.
Our approach
to media development incorporates the classic criteria for media performance
such as optimization of parameters of cell grow rate, maximum
cell density, cell attachment, product yield and component utilization for
the intended application. SAFC animal component free media for biopharmaceutical
applications have been developed to address current and anticipated regulatory
concerns. Our commitment to the development of animal-free media has entailed
re-evaluating the identity and source of all raw materials used in these
formulations. Additionally, plant derived or recombinant forms of essential
growth factors and hormones have been identified. This has also required
the development of synthetic alternatives for such components as cholesterol
and lipids and the development of substitutes for iron transport and lipid
delivery systems. These efforts have lead to the development of animal component-free
media wherin all components can be documented via a Certificate of Origin,
to be isolated from non-animal sources or synthesized using non-animal starting
and processing materials.
