Sample Preparation for Cell Culture Spent Medium Analysis

For most analyses, i.e., amino acids, vitamins, trace elements, etc., a 5 mL sample size for each assay is sufficient.
Cell culture spent medium samples must be prepared prior to testing by separating cells and cellular debris from the medium:
  Harvest cells and media aseptically into 15 mL conical tubes.
  Spin down media at 2600g for 5 minutes. This step insures that no cellular debris interferes with the analysis.
Transfer spent medium supernatant into labeled containers (identified with your company/institution, date, experiment #, and days of incubation) that can be frozen at –20 °C for storage and shipment.
Spent medium samples must be shipped on dry ice.
