General Description Protein sequencing, amino acid analysis, and mass spectrometry of peptides or proteins require treating the samples to minimize concentrations of salts or buffers. Peptides and proteins can be quickly and conveniently separated from interfering salts by using a 2cm x 4.6mm ID Supelguard™ reversed phase guard column filled with a wide pore (300 Angstrom) SUPELCOSIL™ packing. The initial mobile phase, with a high water content, promotes selective binding of the analytes to the stationary phase, while allowing the salts and other small contaminants to pass through the column. A rapid change to a high organic concentration mobile phase elutes the isolated, desalted peptides or proteins. The technique also is effective for concentrating dilute samples. Appropriate Supelguard columns include Supelguard LC-304 (C4 phase, Cat. No. 59591), LC-308 (octyl phase, Cat. No. 59501), and LC-318 (octadecyl phase, Cat. No. 59502).
