Staphylo Monotec Test Kit
Staphylo Monotec test kit (Fluka 08986) is a new rapid agglutination test for differentiation between Staphylococcus aureus and other Staphylococcus (see figure 1). This method has been evaluated for food material and clinical specimens. With this kit coagulase and protein A on S. aureus can be detected in one step. Due to this approach the test is highly sensitive and specific.

Principle
The test reagent consists of monodisperse microparticles which are coated with IgG and fibrinogen. The reagent will detect both cell-associated coagulase and protein A on S. aureus cultured on blood agar or other recommended media for isolation of S. aureus. The negative control reagent consists of monodisperse microparticles coated with bovine serum albumin for use when the agglutination is ambiguous. On average 97% of human S. aureus isolates are coagulase positive. In addition, and independently of the coagulase property, more than 95% of all clinical S. aureus isolates have protein A in the cell wall. Because fibrinogen binds to the cell-associated enzyme coagulase, and because the Fc part of the IgG binds to protein A, a rapid agglutination occurs, when S. aureus is mixed with the Staphylo Monotec test reagent (see figure 2). USE: Bacteria and reagent are mixed for 20 seconds, and results are read immediately. Results are easy to interpret, a positive result gives clear white agglutinates against a black background (see figure 3).

Staphylo Monotec test kit has been evaluated on both human and veterinary/food material [1-2]. The presence of the control reagent is a positive feature which improves the specificity.

b-Lactamase Test Kit
b-Lactams are the most widely used antibiotics, and b-lactamases are the greatest source of resistance to them. Therefore detection and identification of b-lactamase is important.
b-Lactamase Test Kit (Fluka 40561) is a rapid acidimetric test for detection of b-lactamase activity of microorganisms. The test is performed in microtitre wells. This test is based on hydrolysis of the b-lactam ring in benzylpenicillin, which results in the production of penicilloic acid. This process causes acidification of the bacterial suspension, and changes the colour of the acid-based indicator. The acidimetric test for b-lactamase activity is suitable for the detection of Haemophilus influenzae, Neisseria gonorrhoeae and Staphylococcus spp. Not suitable for the detection of the b-lactamase activity of Branhamella catarrhalis, Enteroccocus faecalis, Neisseria meningitis, Enterococcus spp. and others.
