Description

Components

Fluorescein Low Range DNA Standard is provided as two tubes:
• 1 vial (200 μl volume) pBR 322 Hin F1 digest in 10 mM TRIS (pH 8.0), and 1 mM EDTA (50 fmol/μl each fragment; 0.14 μg/μl)
• 1 vial 5× loading buffer (500 μl volume) consisting of 12.5% Ficoll, 0.05% bromophenol blue, 50 mM TRIS (pH 8.0), and 5 mM EDTA

Application

Suitable for size determination of dsDNA by DNA electrophoresis with either agarose or polyacrylamide gels.

General description

Sigma’s Fluorescein Low Range DNA Standard contains 10 fluorescein end-labeled fragments as follows: 75, 154, 220/221, 296, 334, 396, 504, 517, and 1632 bp. The standard should be diluted with the included loading buffer before loading on an agarose or polyacrylamide gel. For detection, the fluorescein should be excited at 302nm for emission at 521 nm.

Other Notes

For optimal resolution, the recommended polyacrylamide gel concentration is < 10% and the optimal agarose gel concentration is > 2.5%, cast < 1mm thick. bp.
