Treatment with diphenyl carbamyl chloride (DPCC) reduces chymotrypsin usually present in trypsin; Thia-analogues of amino acids, synthesis of peptide derivatives containing 3-thia-analogues of amino acids

Unit Definition

1 U corresponds to the amount of enzyme which increases the absorbance at 253 nm by 0.001 per minute at pH 7.6 and 25°C (N-benzoyl-L-arginine ethyl ester, Cat. No. 12880, as substrate)

Application

Tyrpsin is a serine protease used to hydrolyze proteins. Trypsins are used for the re-suspension of cells during cell culture and in proteomics research for the digestion of various proteins . Product 93611 is from bovine pancreas and is DPCC treated. Treatment with diphenyl carbamyl chloride (DPCC) reduces chymotrypsin usually present in trypsin, thia-analogues of amino acids and the synthesis of peptide derivatives containing 3-thia-analogues of amino acids.

Biochem/physiol Actions

Tyrpsin cleaves peptide chains, amides and esters mainly at the carboxyl side of the amino acids lysine or arginine. Tyrpsin plays an important role in digestion and protein absorption. Tyrpsin originates in the pancreas in the form of the zymogen, trypsinogen.
