One unit of Acc I cleaves approximately 98% of the substrate DNA (pBR322 DNA).

Specificity

Recognition Sequence: 5′-GT/(A,C)(G,T)AC-3′
Ligation and recutting results: After 2-10-fold Acc I overdigestion of 1 μg pBR322 DNA substrate, results in 100% cutting, >95% of fragments can be ligated, and >90% recut.
Heat inactivation: Activity retained after 10 minutes at 65 °C. Five-fold enhanced activity occurs at 55°C. Enzyme inactivated at 80 °C for 20 minutes.

Solution in 20 mM Tris-HCl, pH 7.5, 100 mM NaCl, 0.1 mM EDTA, 10 mM 2-mecarptoethanol, 0.01% polydocanol (v/v), 50% glycerol (v/v), at 4°C

AccI is a restriction endonuclease that is used in molecular biological methods to cleave the recognition sequence 5′-GT/(A,C)(G,T)AC-3′, generating DNA fragments with 5′-cohesive termini.
