Supplied with 10x Restriction Endonuclease Buffer SH (B3657).

Comment: Xba I will only partially cleave DNA isolated from E. coli strains that have the dam methylase (dam+ strains). May cleave at secondary recognition sites under non-optimal conditions.

Specificity

Recognition sequence: 5′-T/CTAGA-3′
Ligation and recutting results: After 2-10-fold Xba I overdigestion of 1 μg Ad-2 DNA substrate, results in 100% cutting, >90% of fragments can be ligated, and >95% recut.
Heat inactivation: Up to 15 units of enzyme inactivated at 65 °C for 15 minutes.

Physical form

Solution in 20 mM Tris-HCl, pH 8.0, 1 mM EDTA, 100 mM NaCl, 0.1 mM PMSF, 10 mM 2-mercaptoethanol, 50% glycerol (v/v), 0.02% polydocanol at 4 °C

Application

XbaI is a restriction enxyme that cleaves DNA at the recognition site 5′-T/CTAGA-3′ to generate DNA fragments with 5′-cohesive termini.
