Ubiquitin C-terminal hydrolases (UCHs) are a family of cysteine hydrolases that catalyze the UBPs hydrolyze isopeptide bonds between ubiquitin and the substrate protein or within polyubiquitin chains. Human isopeptidase T (isoT) is an UBP capable of binding ubiquitin that specifically disassembles free polyubiquitin chains linked through isopeptide bonds involving the K48 of ubiquitin into ubiquitin monomers.
