Description

Immunogen

synthetic peptide corresponding to residues 408-439 of the human p62c-Myc protein.

General description

c-Myc is a nuclear protein that modulates cell growth, transformation and cell death. Alterations in c-Myc have been linked to several hematopoietic malignancies such as leukemia and Burkitt lymphoma.
Monoclonal Anti-c-Myc-Alkaline Phosphatase antibody recognizes an epitope located within the sequence EQKLISEEDL (residues 410-419) of the human oncogene product c-Myc. The antibody reacts with both components of the p62c-myc-p64c-myc doublet. The antibody detects human1-11 p62/64c-myc, but does not recognize the chicken p11gag-myc protein present in MC29 virus-transfected quail fibroblasts, nor does it react with the mouse p64/66c-myc protein. Nevertheless, weak reaction with murine c-Myc may be seen when the antibody is used at high concentration.

Physical form

Solution in 0.05 M Tris buffer, containing 1% bovine serum albumin, 1 mM MgCl2, 50% glycerol, and 15 mM sodium azide.

Specificity

Specificity: N-terminal or C-terminal c-Myc (Glu-Gln-Lys-Leu-Ile-Ser-Glu-Glu-Asp-Leu) tagged fusion proteins.

Application

ELISA and dot blot assays using membrane fraction isolated from tumor cells (MDA-MB 231) were performed using alkaline phosphatase-conjugated monoclonal mouse anti-c-Myc antibody. Antibody was developed using p-nitrophenyl phosphate substrate (Sigma).

Monoclonal Anti-c-Myc-Alkaline Phosphatase antibody can be used for western blot assays at 1:100, using myc-tagged fusion proteins. The antibody can also be used for immunohistochemical applications.
