For yeast cells

Frequently Asked Questions

Frequently Asked Questions are available for this Product.

Features and Benefits

• Fast: Cell lysis in just 20 minutes
• Efficient: Three times more efficient than glass beads
• Flexible: Lysis is enhanced by the addition of reducing reagent (DTT)
• Gentle: Non-denaturing - preserves biological activity

General description

CelLytic Y Cell Lysis Reagent is versatile, phosphate-free and non-denaturing. It provides an effective method for cell lysis and protein solubilization. Target proteins maintain immunoreactivity or biological function. The protocol is brief and preformed at room temperature. No extreme conditions or glass beads are required. Use only 2.5-5 ml/gram yeast cells.

Frequently Asked Questions

Which document(s) contains shelf-life or expiration date information for a given product?
If available for a given product, the recommended re-test date or the expiration date can be found on the Certificate of Analysis. These documents are located on the product detail page under Useful Links & Tools. Click on the following link to search for a Certificate of Analysis. Please click the following link to see the details on our Product Dating Information.
How do I get lot-specific information or a Certificate of Analysis?
A Certificate of Analysis is available by lot number and can be obtained through our Advanced Search Option: http://www.sigmaaldrich.com/catalog/AdvancedSearchPage.do
Can I use glass beads with CelLytic™ Y Cell Lysis Reagent, Product C4482?
If cell growth conditions are not optimal, then glass beads can be used to increase yields.  For more information follow this link:  http://www.sigmaaldrich.com/sigma/bulletin/c4482bul.pdf
Should I use CelLytic™ Y Cell Lysis Reagent, Product C4482?
For all cases in which the addition of DTT does not interfere with the application tested, it is recommended to add DTT (5 – 10 mM final concentration) to the CelLytic Y reagent.  Addition of DTT significantly improves total protein yield.
Can I use CelLytic™ Y Cell Lysis Reagent, Product C4482, to make spheroplasts?
No.  For spheroplast formation and protein extraction we recommend the CelLytic Y Plus kit.  The bulletin for CYP1 is here:  http://www.sigmaaldrich.com/etc/medialib/docs/Sigma/Bulletin/c4482bul.Par.0001.File.tmp/c4482bul.pdf
What is in CelLytic™ Y Cell Lysis Reagent, Product C4482?
The formulation is considered proprietary, but we can state that it is a non-denaturing phosphate-free reagent.
What strains have been used with CelLytic™ Y Cell Lysis Reagent, Product C4482?
The efficiency of CelLytic Y reagent for protein extraction has been tested on different strains of Saccharomyces cerevisiae [Y187, Y190, W303(a), S288C, SP1, S1278b (diploid and haploid), BJ2168, and cdc25] and Schizosaccharomyces pombe. More information can be found in the technical bulletin: http://www.sigmaaldrich.com/etc/medialib/docs/Sigma/Bulletin/c4482bul.Par.0001.File.tmp/c4482bul.pdf
