Immunogen

human CCR-5 transfected NSO mouse myeloma cells.

General description

CCR-5 is a chemokine receptor belonging to the super family of seven-transmembrane G-protein coupled receptors that mediates cellular functions of immune response, leukocyte migration, development and angiogenesis. CCR-5 is expressed on macrophages and Th1 cells activated on inflammation, osteoclasts and vascular smooth muscle cells. Three CC chemokines, CCL-3, -4 and -5 bind efficiently to CCR-5, whereas CCL-, -8 and -13 bind less efficiently. The expression of CCR-5 protein is most important in resistance to HIV-1 and hepatitis C virus infections and reportedly influences the course and progression of the infection. Monoclonal Anti-CCR-5 antibody specifically recognizes human CCR-5. The antibody shows no cross reactivity with CCR-1, CCR-2, or CCR-3.

Physical form

Lyophilized from a 0.2 μm filtered solution in phosphate buffered saline with 5% trehalose.

Specificity

The antibody reacts with human CCR-5 transfectant cells and not the parent cell line. The antibody shows no cross-reactivity with CCR-1, CCR-2 or CCR-3. This antibody can be used to detect CCR-5 present on stimulated human PBMCs that have been fixed with 2% formaldehyde, but not on unfixed cells.

Preparation Note

Purified using protein G.

Application

Monoclonal Anti-CCR-5 antibody may be used for flow cytometry; the antibody may be diluted to 25 μg/ml before adding 10 μL of this dilution to 1-2.5 X 106 cells. The antibody detects CCR-5 present on stimulated human PBMCs (peripheral blood mononuclear cells) only after the cells are fixed with 2% formaldehyde. The antibody does not detect CCR-5 on unfixed human PBMCs. For immunohistochemistry a working concentration of 25 μg/ml using paraffin-embedded human lymph node tissue sections.
