Immunogen

Peptide corresponding to amino acids 461-478 of rat CaM Kinase Iiα, conjugated to KLH. This sequence is identical in human, mouse and chicken CaMKIIα has limited homology (50-60%) with CaM Kinase IIβ, γ and δ subunits.

General description

Ca2+/Calmodulin protein kinase II or CaMKII is a serine threonine kinase that regulates various functions such as neurotransmitter secretion, gene expression, spatial learning, axonal transport and long term potentiation through Ca2+ signalling . CaMKII has four different isoforms, namely, α, β, γ and δ . Anti-CaM Kinase IIα specifically binds to CaM Kinase IIα (50kDa) in human, mouse, rat and chicken. CaM Kinase IIα immunizing peptide specifically inhibits the staining of CaM Kinase IIα in immunoblotting. The antibody does not cross-react with CaMKIIβ, γ and δ isoforms (60 kDa).

Intracellular Calcium (Ca2+) acts as major secondary messenger that mediates signalling induced by hormones and neurotransmitters. Cellular responses to calcium are modulated by a family of Ca2+/Calmodulin dependent protein kinases. The members of this family include CaMK I, CAMK II and CAMK IV. These kinases mediate rapid, highly specific and reversible post-translational modification involving ATP. CaMK II is abundantly expressed in mammalian brain, with highest expression in neurons of hippocampus and cerebral cortex, and plays critical role in synaptic transmission, memory, learning and behaviour. CaMK II family has four members, CaMK IIα, CaMK IIβ, CaMK IIγ and CaMK IIδ. The CaMK IIα?and β?isoforms are predominantly expressed in the brain, localized mainly in the cytosol and postsynaptic densities (PSDs), whereas the CaMK IIγ and δ isoforms are expressed in all tissues. CaMK II is autophosphorylated at α-Thr286 residue in response to changes in the concentration of intracellular calcium. This may prolong physiological responses to temporary changes in Ca2+ concentrations. CaMK II regulates synaptic signalling by phosphorylating glutamate receptors in the neurons. A distinct CaMK II has also been implicated in the process of smooth muscle contraction by phosphorylation of myosin light chain kinase.
Anti-CaM Kinase IIα specifically reacts with rat CaM Kinase IIα (50 kDa). This sequence is identical to human, mouse and chicken CaMK IIα.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Specificity

The antibody does not cross-react with CaMKIIβ, γ and δ isoforms (60 kDa).

Application

Anti-CaM Kinase IIα (CaMKIIα) antibody may be used in immunoblotting (50kDa), immunoprecipitation (5 μg using 10 μg of a synaptosomal fraction of rat brain), western blot (1:5,000 using synaptosomal fraction of rat brain) and microarray analyses.

Anti-CaM Kinase IIα (CaMK IIα) antibody detects CaMK IIα in synaptosomal fraction of rat brain by immunoblotting at a working dilution of 1:5000. The antibody may be used for immunoprecipitatiion using 5 μgram in synaptosomal fraction of rat brain. Detection by immunoblotting is possible in cells of mouse hippocampal lysates. . The antibody may also be used for protein microarray.
