General description

Caspase belongs to the family of cysteine proteases and plays crucial roles in extrinsic apoptosis, inflammation, and cellular differentiation. There are at least 14 different caspases with different substrate specificity. Caspase 8 was first identified in humans and at least eight different caspase 8 isoforms have been identified at mRNA level. The predominant caspase 8 isoforms expressed in cells. Caspase 8a/b (479 and 464 a.a.), are formed as inactive 55kDa and 54kDa precursors, respectively. The N-terminal region of caspase 8 contains two death effector domains (DEDs) which facilitate the interaction with the adaptor protein FADD/MORT-1.

The sequence used as the immunogen is identical in human caspase 8 isoforms (MACHα1, MACHα2, MACHβ1, MACHβ3, MACHβ4) and has limited homology (60%) to mouse caspase 8. By immunoblotting, the antibody reacts with caspase 8a and caspase 8b.


Anti-pro-Caspase 8 antibody was used for immunoblotting of protein in a study of a novel short isoform of caspase-8 (caspase-8s), which encodes the first Death Effector Domain (DED) and part of the second DED, missing the C-terminal caspase domain. It is also suitable for microarray and western blotting analysis at a dilution of 1:1,000 by using a whole cell extract from the human Burkitt lymphoma Raji cell line.

Biochem/physiol Actions

The activation of anti-pro-Caspase 8 antibody needs oligomerization-induced processing. It subsequently removes its death domain and cleavage into active 20kD and 10kD subunits which further released in the cytosol. These subunits form a proteolytically active heterodimer which are capable of cleaving other downstream caspase family members.
