The members of epidermal growth factor receptor (EGF R) or the ErbB receptor family have been identified as useful biomarkers and targets for cancer therapy. The EGFR family includes four receptor tyrosine kinases, EGF R (ErbB1), ErbB2 (Neu), ErbB3, and ErbB4. EGF R binds EGF and induces tyrosine phosphorylation leading to proliferation of cells. 3,4 EGF R is present on many cell types of epithelial and mesenchymal lineages. EGF R is capable of binding transforming growth factor-α and heparin-binding EGF in addition to EGF. 5,6 There are numerous effector molecule activated by EGF R that result in a variety of biological processes such as morphogenesis, cell motility, apoptosis, differentiation and organ repair and maintenance. Deregulation of EGF R signaling is implicated in progression of a wide variety of tumors, invasion and metastasis3,6. Monoclonal anti-EGF Receptor detects human, mouse, pig, rat and canine EGF receptor.
Localizes blood type A-related determinants on various glycoproteins and glycolipids in addition to specific EGF receptor determinants (only observed in human tissue). Binds an external carbohydrate on the receptor and does not block EGF binding.
Monoclonal Anti-Epidermal Growth Factor Receptor antibody may be used for ELISA at a working antibody dilution of 1:10000 using the extract of A431 cells. For immunoblotting, a dilution of 1:3000 using A431 cell extract is recommended. The antibody may be used to purify EGF receptor, for structural and functional studies of the receptor and for investigation of cell transformation versus oncogenesis. Antibody dilution of 1:50 was used for immunohistochemistry of rat whole ovary sections.1 The antibody was also used for immunoprecipitation of EGF receptor from extracts of A431 cells.2
