Description

Preparation Note

Prepared using mild extraction conditions to minimize peptide bond cleavage as described by Starcher, et al.

Application

Elastin is a structural protein which has been used in studies of Chronic obstructive pulmonary disease (COPD). It may be used to investigate why there is a lack of repair for these proteins in patients with COPD.

Biochem/physiol Actions

The cyclooxygenase-2 inhibitor rofecoxib binds covalently to the modified amino acid allysine in human aorta elastin, blocking cross-linking and causing degradation of elastic fibers. Other structurally similar COX-2 inhibitors do not show the same activity. Nevertheless, this activity is postulated to be responsible for adverse cardiovascular events in COX-2 administration.
