pFLAG-CMV™-3 is a shuttle vector for E. coli and mammalian cells, containing both bacterial and SV40 origins of replication. It is used for transient or stable expression and secretion of N-terminal FLAG®-tagged fusion proteins in mammalian cells. Efficiency of replication and genomic integration is optimal in mammalian cells that express the SV40 large T antigen, such as COS cells. The promoter-regulatory region of human cytomegalovirus drives expression of the FLAG®-tagged fusion constructs that contain a preprotrypsin leader sequence preceding the FLAG® sequence in order to direct secretion into the culture medium. It also contains the aminoglycoside phosphotransferase II gene, which confers resistance to aminoglycosides such as neomycin (G 418), allowing selection of stable transformants. It has been used successfully for stable transfection of HEK 293 cells.
