Components

Cellular fibronectin is an adhesion glycoprotein of the ECM,k existing as a dimer of an A chain and the B chain containing the type III connecting segment (IIIcs) region. Cellular fibronectin is different from plasma fibronectin by the presence of additional polypeptide segments and in an altered morphology of transformed cells and hemagglutination. Different forms of fibronectin can be generated from tissue specific splicing of fibronectin mRNA transcribed from a single gene.

Analysis Note

Protein homogeneity is evaluated by immunoelectrophoresis.

Application

This product is recommended for use as a cell culture substratum at 1-5 μg/cm2 or 0.5-50 μg/mL. The optimal concentration does depend on cell type as well as the application and research objectives. Bovine plasma fibronectin has been shown to stimulate the proliferation of rat osteoblasts in vitro, to induce odontoblast polarization and to regulae the differentiation of human dental pulp cells into odontoblasts.

Biochem/physiol Actions

Domains of fibronectin bind with collagen, fibrin, heparin, and specific cell membrane receptors. The most notable domain, Arg-Gly-Asp can be recognized by integrins and mediates cell adhesion. Fibronectin is also widely involved in interactions such as the attachment and migration of many cell types, cytoskeletal assembly, tyrosine phosphorylation, and metastasis.

How do I choose which fibronectin product to use to coat culture surfaces for cell growth?
The species source for the fibronectin used when coating culture surfaces is not important.  It is the presence of the RGD binding site that is present in all fibronectins that promotes binding.
How long can I store the fibronectin solution?
The product is good for at least a year at 2-8 °C if kept sterile.
How long can I store fibronectin coated plates, etc?
Fibronectin coated cultureware can be stored for 2-4 weeks at 2-8 °C in a closed sterile container, or in sterile sealable bags.
What is the procedure for coating culture surfaces with fibronectin?
To coat culture surfaces: 1. Dilute fibronectin in sterile balanced salt solution and coat the culture surface (1-5 μg/cm2) with a minimal volume.  2. Allow to air dry for at least 45 minutes at room temperature.  Excess fibronectin may be removed by aspiration, but this is not necessary.
