Description

Immunogen

recombinant soluble active extracellular domain of human Fas ligand.

General description

The Fas ligand (Fas ligand, FasL, CD95L) is an integral membrane protein and is a type II transmembrane glycoprotein expressed by CD4+ and CD8+ cytotoxic T lymphocytes, natural killer and a small number of non-lymphoid cells. FasL is a member of the TNF family. Membrane bound FasL (mFasL) is a 40kDa protein, while the active soluble form of FasL (sFasL) was identified as a 26kDa protein. FasL has four potential N-glycosylation sites.

Preparation Note

Purified from culture supernatant of hybridoma cells grown in a bioreactor

Application

Monoclonal Anti-Fas Ligand antibody is suitable for flow cytometry at a concentration of 1-2μg/test using cultured KFL9 cells (human chronic myelogenous leukemia cell line K562, stably-transfected with human Fas Ligand). It is also suitable for immunocytochemistry; immunoprecipitation; indirect ELISA, microarray analysis.

Biochem/physiol Actions

Fas ligand (FasL) binds to the Fas receptor and triggers apoptosis, which plays a crucial role in the maintenance of immune system homeostasis. Fas/FasL play crucial role in carcinogenesis, tumor outgrowth, and metastasis. Mutation in FasL is associated with systemic lupus erythematosus.

Antibodies to Fas (CD95/Apo-1) and Fas Ligand (CD95L)

Many cells can be activated to undergo apoptosis following the interaction of selected ligands with cell surface receptors. The most well studied receptors are CD95/Fas/Apo-1 (apoptosis inducing prot...
