Description

Immunogen

synthetic peptide corresponding to amino acids 725-741 of human GIT1, conjugated to KLH. The corresponding sequence is identical in human GIT1 isoforms 1and 2, and in rat and mouse GIT1.

General description

GIT1 (G protein-coupled receptor kinase interactor 1) is an adaptor protein with GTPase-activating protein activity for the ADP-ribosylating factor. It is also named as Cat-1, p95-APP1 and p95PKL.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Application

Anti-GIT1 (C-terminal) antibody is suitable for immunoprecipitation at a concentration of 15-20μg using rat brain extract (S1 fraction) and western blot at a concentration of 1-2μg/mL using rat brain extracts (S1 fraction) and mouse brain (S1 fraction) extracts.

Biochem/physiol Actions

GIT1 (G protein-coupled receptor kinase interactor 1) is predominantly distributed and exists in its full length form. It helps to control cellular responses through the activation of heterotrimeric G proteins. GITs can interact with several proteins including GRKs, ARF1, Rac1, Cdc42, PAK, MEK1, PLCg1, FAK, paxillin and PAK-exchange-interacting factor (PIX). It is also involved in receptor internalization. It helps in regulating membrane trafficking between the plasma membrane and endosomes, targeting ARFs to the ARF-GAP activity of GITs. Also it has been found to play a pathogenic role in Huntington′s disease.
