Description

Biochem/physiol Actions

Gelsolin is an actin-severing protein found in mammalian cells and blood plasma. Exhibits tumor suppressor activity in several cancer cell lines and in nude mice.

Gelsolin is an actin-binding protein that regulates the assembly and disassembly of actin filaments.

Unit Definition

One unit will reduce viscosity difference between actin solution and buffer by 50% in a 1 mL mixture containing 1-2 mg F-actin, 0.15 M KCl, 20 mM Tris, pH 7.6, 0.2 mM CaCl2, 0.2 mM ATP and 1 mM DTT at 28°C.

Physical form

Lyophilized powder containing Tris buffer salts, pH 7.6, NaCl, DTT, and EGTA

Application

Plasma gelsolin levels may be used to identify patients at risk for increased morbidity.

General description

Human gelsolin (also known as brevin) is an actin severing protein found in mammalian cells and blood plasma. Gelsolins consist of a 730 amino acid core structure which contains six repeated domains. These six domains make up three distinct actin binding sites. The plasma enzyme contains an N-terminal extension of 25 amino acids that raises its molecular weight from 90 kD to 93 kD.
