Opiorphin is a natural analgesic, secreted into human saliva. Opiorphin demonstrates dual-inhibitory potency on the enkephalin-inactivating ectopeptidases human NEP (hNEP) and human AP-N (hAP-N). As an analgesic, Opiorphin is six times stronger than the opiate morphine. Opiorphin is closely related to the rat sialorphin peptide, which is an inhibitor of pain perception and acts by potentiating endogenous micro- and δ-opioid receptor-dependent enkephalinergic pathways. In rat studies, Opiorphin suppressed pain sensation for both chemical-induced inflammation and acute physical pain. In both cases, the administered dose of 1 mg/kg provided the same painkilling power as 3-6 mg/kg of morphine.
