Specificity

Recognition sequence: 5′-CCGC/GG-3′
Ligation and recutting results: After 2-10-fold Ksp I overdigestion of 1 μg λ DNA substrate, results in 100% cutting, >90% of fragments can be ligated, and >90% recut.

Solution in 20 mM Tris-HCl, pH 7.5, 0.1 mM EDTA, 100 mM NaCl, 10 mM 2-mercaptoethanol, 50% glycerol (v/v), 0.01% gelatine (v/v), 0.2% polydocanol (v/v) at 4°C

Application

KspI is a restriction endonuclease that is used for molecular biology applications to cut DNA at the recognition sequence 5′-CCGC/GG-3′, generating DNA fragments with 3′-cohesive ends.
