Description

Immunogen

synthetic peptide corresponding to amino acid residues 163-177 of human Rab7 with C-terminal added cysteine, conjugated to KLH.

General description

Anti- Rab7 antibody may be used for immunoblotting at a working concentration of 2.5-5.0 μg/ml using whole cell extracts of human A431 cells. For immunofluorescence staing of rat NRK and mouse NIH3TS cells, a working concentration of 5-10 μg/ml is recommended. Anti-Rab7 was used for immunofluorescence using human fetal foreskin fibroblasts (HFFF-2).

Rab7 belongs to Ras superfamily of small GTPases. It facilitates the maturation of late autophagic vacuoles and regulates vesicle trafficking from early to late endosomes and also from late endosomes to lysosomes. Anti-Rab7 antibody can be used to study subcellular localization and membrane organization. It can also be used as a marker for late endosomes. Rabbit anti-Rab7 antibody reacts specifically with Rab 7 from mouse, rat and human.

Rab7 belongs to the GTPases superfamily of small molecular weight proteins. It is a key regulator of late endosome trafficking and promotes EGFR trafficking from cell surface to lysosomes. The molecular functions of Rab7 range from regulating the cargo movement out of early endosomes to lysosomal delivery from late endosome/multivesicular body. The role of Rab7 is essential for the degradation of EGF-EGFR complex in the lysosome. Rab7 is crucial in maintaining cellular homeostasis, intracellular transport, uptake of nutrients, immune response and signalling through receptors on the cell surface
Anti- Rab7 recognizes human, mouse and rat Rab7 (~23 kDa).

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Application

Anti-Rab7 antibody can be used in indirect immunofluorescence and immunoblotting. It can also be used in western blot.
