TBB is a highly selective, ATP/GTP-competitive inhibitor of casein kinase-2 (CK2) (IC50 = 900 nM and 1.6 mM, using rat liver and recombinant human CK2, respectively).
TBB binds to the Val66 residue of casein kinase-2 and inhibits the binding of ATP/GTP.2 TBB is cell permeable; it induces caspase-dependent apoptosis and degrades hematopoietic lineage cell-specific protein 1 in Jurkat cells.3
TBB was used to study casein kinase 2-dependent phosphorylation of DNA damage mediator protein MDC1.1
