Application
These cells have been derived from a patient with erythroleukemia. They show complete growth dependency on GM-CSF or IL-3, and carry a homogenious chromosomal abnormality (54,X). Constitutive expression of globin genes indicate concommitment of TF1 to the erythroid lineage, although glycophorin A or carbonyl anhydrase I are not expressed. On incubation with hemin or δ-aminolevulinic acid hemoglobin synthesis was induced, whereas TPA induced differentiation into macrophage-like cells. Erythroprotein also sustains short-term growth, but does not induce differentiation.
Maintain cultures at 2-9 × 105 cells/ml. Incubate at 37 oC and 5% CO2.
Recommended culture medium: RPMI-1640 + 2 mM L-Glutamine + 1% Sodium pyruvate + 2-5 ng/ml Human GM-CSF + 10% FBS. Replenish GM-CSF every 48 hours.
