Reconstitution
Dilute 10× Blocking Buffer 1:10 with deionized water prior to use (e.g. Add 1 mL of 10× Blocking Buffer and 9 mL deionized water to obtain a 1× working concentration).
Application
Blotting applications, using colorimetric or chemiluminescent detection, require the use of an added blocking step to decrease non-specific signal generated by non-specific binding of enzyme conjugates to the membrane. Sigma has developed 10× Blocking Buffer, a casein-based buffer, to provide the highest signal-to-noise performance over blocking buffers containing dried milk or bovine serum albumin (BSA). 10× Blocking Buffer eliminates the inconvenience associated with preparation of blocking buffers containing caseins which are difficult to dissolve; it is supplied as a 10× concentrate and only requires dilution with distilled water prior to use. The 10× Blocking Buffer is versatile and functions with neutral and positively-charged nylon membranes (See BioBond™ and BioBond™ Plus nylon membranes). 10× Blocking Buffer is recommended for both colorimetric and chemiluminescent detection in membrane-based blotting applications.
