General description
GC5™ competent Escherichia coli is a high transformation efficiency strain that is comparable to DH5α™. GC5 chemically competent cells are suitable for molecular biology applications such as plasmid propagation, cDNA library generation from plasmid-based vectors, and general cloning protocols. This K strain bacterium carries the recA1 mutation to minimize recombination and provide plasmid stability and the endA1 mutation to improve the quality of plasmid DNA preparation. GC5s carry the Δ(lacZ)M15 mutation for blue/white screening of recombinants via α-complementation with the amino terminus of β-galactosidase. In addition, the tonA genotype confers resistance to the lytic bacteriophages T1 and T5 for protection of clonal stocks.
Sigma's GC5 competent E. coli cells are grown and made chemically competent using an optimized procedure specific to the strain, followed by strain verification and efficiency testing. The cells are provided in a 96-well plate (20 μl/well) for high throughput transformations. Each well may be used for a single transformation. The cells have a transformation efficiency of >1x108 cfu/μg when transformed with non-saturating amounts of pUC19 plasmid DNA.
