Its primary site of action appears to be on neurons in the hypothalamus that are involved in regulating energy balance, appetite, and body weight. Leptin increases the production of nitric oxide in endothelial cells and stimulates angiogenesis in vitro and in vivo.
Human and mouse leptin share ~84% sequence identity.

OB-R, also known as B219, is expressed in the yolk sac, early fetal liver, enriched hematopoitic stem cells and in a variety of lymphohematopoietic cell lines. OB-R is also expressed at high levels in adult reproductive organs. OB-RL action in hepatic cells may be equivalent to that of IL-6 receptor. Leptin does not play a specific role in muting insulin action on hepatoma cells and may not contribute to the diabetic symptoms associated with obesity.
