Trizma® (TRIS base)

Synonym: 2-Amino-2-(hydroxymethyl)-1,3-propanediol, THAM, Tris base, Tris(hydroxymethyl)aminomethane, Trometamol
 

TRIS is an established basimetric standard and buffer used in biochemistry and molecular biology1. It is used in the formulation of buffer solutions in the pH range between 7.5 and 8.5. TRIS buffer solutions are widely used in cell and molecular biology for processes such as protein and nucleic acid extraction and purification.
Useful pH Range 7 - 9
Working concentration range 10-62.5mM9,10

Interaction Comments
 

TRIS may form Schiff bases with aldehydes and ketones.5
TRIS has been reported to interfere with the activity of a number of enzymes, it should therefore be used carefully when studying proteins2,3.
TRIS exhibits a large shift in dissociation with a change in temperature.6
TRIS interferes with the Bradford dye-binding protein assay4
Strong interaction with Cr, Fe, Co, Ni and Cu; weak interaction with Mg, Ca, Zn, Cd and Pb8
