The MISSION siRNA Transfection Reagent is for the transfection of small RNA (siRNA & miRNA) to achieve ≥90% silencing efficiency for transient knockdown of eukaryotic gene expression. Effective gene silencing and reduced off target effects are achieved using a low siRNA concentration (1nM). The siRNA Transfection Reagent is provided as a sterile solution, and is compatible with serum and antibiotics.

Benefits

  • Superior gene silencing in a wide variety of eukaryotic cell types
  • Effective transfection with low RNA concentrations, reducing off target effects
  • Use in full-serum, serum-free media and with antibiotics
  • Excellent cell viability after transfection
  • For use with siRNA, esiRNA, miRNA mimics and Synthetic miRNA inhibitors

Figure 1a. Cell lines and cells that have been successfully transfected with siRNA, and the silencing efficiencies for each using the MISSION siRNA Transfection Reagent.

Figure 1b. Percent inhibition of four adherent cell lines measured 48 hours after transfection with1 nM siRNA against GADPH, using the MISSION siRNA Transfection Reagent.

High silencing efficiency

Figure 2. 3T3 cells were transfected with 5 nM siRNA (final concentration) against Vimentin (Left), and a siRNA negative control (Right) using the MISSION siRNA Transfection Reagent. Immunofluorescence was performed 48 hours after transfection using a primary antibody against Vimentin and a fluorescently-labeled secondary antibody.

Reduced cytotoxicity

Figure 3. A549 cells stably expressing GL3 Luciferase were transfected with 1nmol siRNA against luciferase. Cell morphology was compared after 48 hours using the MISSION siRNA Transfection Reagent (Left) and another manufacturer’s transfection reagent (Right).
