HPLC Analysis of Lysozyme on SRT® SEC-300, 30 cm x 7.8 mm I.D., 5 μm particles

Conditions

column SRT SEC-300, 30 cm x 7.8 mm I.D., 5 µm particles (Z777051)
column temp. ambient
mobile phase 50 mM phosphate buffered saline, 200 mM Na2SO4, pH 7.0
flow rate 1 ml/min
sample 1 mg/mL in 50 mM phosphate buffered saline
injection 5 µl
detector UV, 214 nm

Description

Analysis Note The positively-charged protein, lyzoyme (pI=11), exhibits excellent peak shape and recovery on SRT SEC-300 columns due to its highly-inert surface. Low protein recovery is usually attributed to 1) hydrophobic interactions with the stationary phase and 2) electrostatic interactions with the silica suface. SRT SEC-300 surface is hydrophilic and densely bonded to silica, therefore providing high mass recovery and good peak shape.
