Elastic Stain

Reagent Preparation

Working Elastic Stain solution is prepared by adding (in order):
Hematoxylin solution (20 mL); Ferric Chloride solution (3 mL);
Weigert’s Iodine solution (8 mL); & Deionized Water (5 mL).
Mix well.
Differentiation: Working Ferric Chloride solution is prepared by adding 3 mL of Ferric Chloride solution to 37 mL deionized water. Prepare working solutions fresh daily.

Procedure

  1. Deparaffinize tissue sections and hydrate to deionized water.
  2. Place in working Elastic Stain for 10 minutes.
  3. Rinse in deionized water.
  4. Differentiate slides individually in working Ferric Chloride solution.
  5. Rinse in tap water.
  6. Check microscopically. If overdifferentiated, return to Working Elastic Stain solution.
  7. Rinse in 95% alcohol to remove iodine.
  8. Rinse in deionized water.
  9. Stain in Van Gieson solution for 1-3 minutes.
  10. Rinse in 95% alcohol.
  11. Dehydrate in alcohol, clear in xylene and mount.

Results

Elastic Fibers – Blue black to black
Nuclei – Blue to black
Collagen – Red
Muscle and Other – Yellow
