Hematoxylin Stain–Mayer

Procedure

  1. Deparaffinize tissue sections and hydrate to deionized water.
  2. Place slides in Mayer’s Hematoxylin solution for 15 minutes.
  3. Rinse in warm running tap water for 5 to 15 minutes.
  4. Rinse in deionized water for 30 seconds.
  5. Stain slides in Eosin Y Alcoholic for 30 seconds.
  6. Dehydrate in alcohol, clear in xylene and mount.

Nuclear Staining for Special Stains

  1. Complete individual staining procedure.
  2. Rinse in deionized water.
  3. Stain slides in Mayer’s Hematoxylin solution for 3-5 minutes.
  4. Rinse in running tap water until nuclei are blue.
  5. Dehydrate in alcohol, clear in xylene and mount.

Results

Nuclear chromatin should be blue. Nucleoli should be conspicuous and crisply outlined. Cytoplasm will display various shades of pink and red blood cells will be red.
