Recipes for Western Blotting Buffers & Stock Solutions

Prepare your solutions accurately, whether you are mixing one liter for yourself, or 50 liters for the lab. Choose the buffer you wish to prepare from the list below. Solutions for Sample Preparation, Blocking, & Immunodetection

4X SDS-PAGE sample loading buffer (order premixed)
 

  7.5 mL Recipe Desired Volume: mL 
  3 mL of 1 M DTT (dithiothreitol) (D9779)
Use mL of 1 M DTT (dithiothreitol)
  0.6 g of SDS (sodium dodecyl sulfate) (L3771) Use g of SDS (sodium dodecyl sulfate)
  0.03 g of bromophenol blue (B0126) Use g of bromophenol blue
  2.4 mL of glycerol (G5516) Use mL of glycerol
  Bring final volume to 7.5 mL with Milli-Q® Water Bring final volume to   mL with Milli-Q® Water
     
  • If solution is orange/yellow in color, add 1 drop of 5 M NaOH to adjust pH
  • Make 500 µL aliquots and store at –20°C

TBS 10x (concentrated Tris-buffered saline) (order TBS powder or tablets)
 

  1 L Recipe Desired Volume:
  88 g NaCl (formula weight: 58.4 g) (S3014)
Use g NaCl
  24 g Tris base (formula weight: 121.1 g) (T1503) Use g Tris base
  Dissolve in 900 mL Milli-Q® Water Use L Milli-Q® Water
  pH to 7.6 with 12 N HCl (H1758) pH to 7.6 with 12 N HCl
  Add Milli-Q® Water to a final volume of 1 L Add Milli-Q® Water to a final volume of L
     

1X solution: mix 1 part of the 10X solution with 9 parts Milli-Q® water and adjust pH to 7.6.

TBST (Tris-buffered saline, 0.1% TWEEN® 20) (order TBS powder or tablets)
