Doxycycline hyclate is a semisynthetic tetracycline functioning as a broad spectrum antibiotic and bacteriostatic agent effective against Gram-positive and Gram-negative bacteria. The recommended usage concentration in cell culture applications is 0.1-12 ug/ml.

Doxycycline hyclate is applied in tetracycline (doxycycline)-inducible gene expression as a stable switch for controlling transcriptional activation, also known as a Tet-On/Tet-Off system. Doxycycline-controlled gene expression is not limited to mammalian expression; it has been adapted for use in a variety of microorganisms, including fungi, parasites and bacteria. Doxycycline is increasingly being utilized in transgene animal model and cell line development, as well as inducible shRNA expression for RNAi mediated gene knockdown. It is easily administered via water supply to animal models for in vivo studies.

At subantimicrobial doses, doxycycline functions as a matrix metalloproteinase (MMP) inhibitor and induces apoptosis through mitochondrial mediated pathway in various tissues. It has recently been identified with having anti-cancer properties.
